چکیده

Antimicrobial peptides (AMPs) play an important role in innate immunity against pathogenic infections. AMPs exterminate bacteria by disrupting cell membranes or inhibiting intracellular molecules. NK-2, first identified pigs and derived from NK-lysin, has antimicrobial effects parasites. In this study, chimeric (cpNK) of chicken pig NK-2 cpNK-derived (cpNK-a1 cpNK-a2) were synthesized, their various such as Escherichia coli, Salmonella spp., Listeria monocytogenes, Staphylococcus aureus, methicillin-resistant aureus (MRSA) investigated. The structure cpNK, include α-helix like peptide net charge was +9 porcine NK-2. cpNK showed powerful bactericidal most bacterial species, including MRSA, especially gram-negative bacteria. Furthermore, short peptides, cpNK-a1 a2 also activity, but the weaker than those cpNK. Therefore, we conclude that cpNK- have potential to be used antibiotic alternatives.

Antimicrobial activity of chicken NK-lysin against Eimeria sporozoites.

NK-lysin is an antimicrobial and antitumor polypeptide that is considered to play an important role in innate immunity. Chicken NK-lysin is a member of the saposin-like protein family and exhibits potent antitumor cell activity. To evaluate the antimicrobial properties of chicken NK-lysin, we examined its ability to reduce the viability of various bacterial strains and two species of Eimeria pa...

The antimicrobial peptide NK-2, the core region of mammalian NK-lysin, kills intraerythrocytic Plasmodium falciparum.

In a time of dramatically increasing resistance of microbes to all kinds of antibiotics, natural antimicrobial peptides and synthetic analogs thereof have emerged as compounds with potentially significant therapeutical applications against human pathogens. Only very few of these peptide antibiotics have been tested against protozoan pathogens that are a major cause of morbidity and mortality in...

Membrane active antitumor activity of NK-18, a mammalian NK-lysin-derived cationic antimicrobial peptide.

As the increasing emergence of multi-drug resistant tumor cells, there is an urgent need for developing new chemotherapeutic agents. NK-lysin was a novel effector of cytotoxic T cells and natural killer (NK) cells and had broad antimicrobial activity. In this study, we developed a core region of NK-lysin termed NK-18, and studied its antitumor activity and possible action mode. Our results show...

Effects of a single nucleotide polymorphism in the chicken NK-lysin gene on antimicrobial activity and cytotoxicity of cancer cells.

NK-lysin is an effector protein of the innate immune system and an important component of host protection. We isolated a SNP in the NK-lysin coding sequence among different chicken breeds. This A to G substitution at the position 271 nucleotide in the ORF results in an Asn (N) to Asp (D) amino acid alteration. We synthesized two 30-aa peptides (N29N and N29D) to compare the biological activity ...

Characterization of a NK-lysin antimicrobial peptide gene from channel catfish.

Natural killer (NK) cells and cytotoxic T lymphocytes (CTL) are important and powerful components of mammalian host defenses against a broad range of microbial assaults. These cell types, upon stimulation, are known to release cytolytic granules toward target cells (reviewed in [1,2]). These granules contain proteins, including perforin and the granzyme family, capable of causing membrane pertu...
